Roanoke County Sheriff's Sgt. Roger Robinson said the two vehicles hit head-on shortly after 8 p.m.
It took rescue workers more than an hour to remove the two men from the car, which was broken in half by the impact.
The accident occurred near Apple Grove Lane.
The identities of the two men had not been released by 10 p.m. because their families had not been notified.
The accident happened as the car came out of a sharp curve near a 35-mph zone. Robinson said the tractor-trailer driver tried to avoid the collision. Witnesses said they heard screeching brakes before the accident, which sounded like "an explosion."
Robinson said the truck driver suffered minor injuries.
