Eldin Paulino hit a two-run home run off Salem's Stan Fansler (0-4), and the Spirits got another run from an error to take a 3-0 lead in the third inning.
In the fifth, John Jensen doubled off the wall to drive in another Spirits run.
Salem (5-8) scored in the seventh when Greg Sims singled to left center with one out, moved to second on a wild pitch and scored from second on a single by Mandy Romero.
Winston-Salem (10-3) got two more in the eighth when Paulino hit a two-run double off Chip Duncan.
Lopez, who was 18-8 last year in Class A Peoria, Ill., of the Midwest League, pitched a complete game in his second start in the Carolina League since being demoted from Class AA Charlotte, N.C., of the Southern League this year.
Winston-Salem puts its seven-game winning streak on the line as the series continues tonight.
