While a statement made public Saturday by the vice president's office did not specifically disclose the ailment, other Bush administration officials said it was cancer.
The statement said Marilyn Quayle, who will be 41 next week, was "recovering well" and "will be able to resume a full schedule in four to six weeks."
The surgery has been planned for several weeks, and diagnosis for the cancer was made two months ago, said officials.
