With the triumph, the Highlanders (19-6) recorded the second most victories in school history.
"We played well in another tough road game," said Radford coach Oliver Purnell. "We held our own and had enough of a spurt at the end to pull it out."
Radford used a 9-2 run at the start of the second half to seize a 40-25 lead at the 17:09 mark, but almost just as quickly the College of Charleston (13-10) erased the deficit.
The Cougars scored the next 13 points, and eventually took the lead 44-43 on a hoop by Mike Beckett, who scored a game-high 16. The lead see-sawed after that.
Then with 5:09 remaining, Tyrone Travis put Radford up 51-50 with a stick-back basket. That was the start of a 6-0 spurt that gave the Highlanders a 55-50 advantage with 1:05 left. Radford missed four free throws in the final minute, but the College of Charleston got no closer than three.
Chris Hawkins added 11 points for Radford, and Travis had a game-high 11 rebounds.
