"The new store will better serve our customers and our donors with a more accessible, more modern shopping environment," said John Wade, the agency's executive director.
The additional space at the new facility means there will be a greater need for donations of clothing and household items, Wade added.
Revenue from the store helps fund the agency's vocational rehabilitation programs.
Donations will be accepted at the old store, 3125 Salem Turnpike, from 8 a.m. to 4:30 p.m. Monday through Saturday and 9:30 a.m. to 4:30 p.m. Sunday.
The Tinker Mountain Sheltered Employment Center on U.S. 11 in Troutville will accept donations from 8:30 a.m. to 4:30 p.m. Monday though Friday.
The new store will accept donations on weekends through mid-April.
