It was the second women's lacrosse title in three years for Virginia, which won its last nine games to finish 15-1.
Top-ranked UVa, a 15-7 winner over Princeton in the regular season, fell behind 4-1 and needed a goal by Anna Yates with 16:07 remaining to send the game into overtime at 5-5.
After Princeton had gone ahead 6-5 in the first of two three-minute overtimes, Kelly Daddona scored with 39 seconds remaining to make it 6-6.
Virginia pulled away in the second overtime on goals by Kelly Cowan and Cherie Greer. Cowan and Heather Goehringer had two goals apiece to lead UVa.
The Cavaliers used two rain and lightning delays to regroup earlier.
"It was a good delay for us," coach Jane Miller said. "We made one major adjustment by putting Heather Goehringer back taking the draw and that gave us a deep breath."
The Cavaliers reached the championship game with an 11-10 overtime victory Saturday over Harvard in a game they twice trailed by three goals.
It was the second NCAA championship of the year for UVa, which earlier captured the men's soccer title for the third time in four years. ACC teams have won the past three women's lacrosse championships, including Maryland in 1992.
