Alyson Whitaker, 16, the driver, was taken to Baptist Hospital-Bowman Gray School of Medicine in North Carolina for treatment of spinal injuries, state police said.
She was listed in serious condition in the intensive care unit and has a spinal fracture, a spokesman said.
Two passengers, Lorrie Felty and Lindsey Graham, also were injured and were flown to Roanoke Memorial Hospital.
The accident happened about 5 p.m. Saturday on Jennelle Road in Montgomery County, 2.3 miles off U.S. 460 near Blacksburg.
Trooper H.A. Spahr said as the car approached a curve, it ran off the right side of the road, plunged down an embankment into some large rocks, then overturned.
Felty, who turns 17 Friday, was able to get out of the back seat and go to a nearby house to call for help, police said. Felty was in satisfactory condition, a nursing supervisor said Wednesday afternoon. She has a broken wrist and a broken elbow, according to Stanley Bragg, band director at the high school.
Graham, 16, a passenger in the front seat, broke her jaw in three places, Bragg said. The hospital had no information Wednesday about her condition.
Bragg said the three girls, all rising juniors, had been planning to leave Sunday for a band camp at Ferrum College.
Bragg said a bake sale has been organized to raise money for Whitaker's medical expenses. It will be held Saturday at Kroger in Spartan Square, Salem, 7 a.m.-2 p.m. Those wishing to donate items for the sale may call 268-2542.
