Barker hit an 0-2 pitch off Brandon Parker down the left-field line to make a winner of Tech starter Brian Fitzgerald (7-5), who allowed just two hits over the first five innings.
The top-seeded Hokies (34-22) took a 2-0 lead when Barker doubled home a run in the first inning and Matt Reynolds scored on a squeeze bunt in the fourth.
Southern Miss tied the score with a pair of runs in the sixth inning, setting up Barker's heroics.
Tech faces second-seeded South Florida today at 7:30 p.m.

Brian Sharp of Virginia Tech shot a 3-over-par 73 in the first round of the NCAA Division I East Regional golf tournament at Yale University in New Haven, Conn.
Sharp, playing as an individual, is tied for 37th going into today's second round.
Clemson, paced by individual leader Mike Byce (67), leads the team competition with a 282 aggregate, two strokes better than North Carolina.
The top 11 teams and the next two low individuals will advance to the NCAA championships.
Two executives of the NFL expansion Carolina Panthers will be the guests at the final dinner-meeting for the 1994-95 Roanoke Valley Sports Club year on Tuesday, May 30 at the Salem Civic Center. Mark Richardson, director of business operations, and Charlie Dayton, director of communications and broadcasting, will discuss the new NFL franchise.
