Barker hit an 0-2 pitch off Brandon Parker down the left-field line to make a winner of Tech starter Brian Fitzgerald (7-5), who allowed just two hits over the first five innings.
The top-seeded Hokies (34-22) took a 2-0 lead when Barker doubled home a run in the first inning and Matt Reynolds scored on a squeeze bunt in the fourth.
Southern Miss tied the score with a pair of runs in the sixth inning, setting up Barker's heroics.
Tech faces second-seeded South Florida today at 7:30 p.m.
