The accident occurred about 4 p.m. when a tractor-trailer in the left southbound lane decided to change lanes, state Trooper M.D. Austin said.
As the truck moved into the right lane, it hit a car, causing its driver to lose control. The car crossed the median and was hit by a northbound tractor-trailer.
Austin said the truck that caused the accident continued down the highway.
The accident is being investigated as a hit-and-run; Wednesday night, no arrests had been made.
The car's driver and a passenger were airlifted to Roanoke Memorial Hospital.
Austin did not know the condition of the passenger. He would not release the names or genders of either victim.
The driver of the second tractor-trailer was not injured, Austin said.
