The victims were identified as James and Johnny Holland, said Claude Webster, Franklin County's director of public safety.
Webster said the exact ages of the men had not been determined, but both were in their late 40s.
The brothers lived in a singlewide mobile home in a hollow off U.S. 220 just north of Boones Mill.
Webster said the trailer caught fire when a small portable heater ignited some combustible material - including carpet - that was too close to it.
Firefighters arrived about 4:45 to find the dwelling engulfed in flames. After knocking down the fire with water hoses, the firefighters entered the trailer and discovered the bodies, according to Webster.
Autopsies are being done on the brothers, but both are believed to have died from smoke inhalation, Webster said.
There is no evidence of foul play, he said.
